Abstract of A STUDY OF PRG4 AS IT RELATES TO URIC ACID AND GOUT, by Talia D’Ambruoso, ScM, Brown University, DECEMBER, 2021. Objective: Proteoglycan-4 (PRG4) is a mucinous glycoprotein expressed by synovial fibroblasts and plays an important role in joint heath. The objective of thesis was to identify a new mechanism of action by PRG4 as it relates to gout and uric acid. Methods: MSU crystal growth studies were conducted comparing rhPRG4 to bovine submaxillary mucin (BSM) as a comparator., CXCL and IL1β ELISA studies in macrophages from Prg4-/- mice, and conditional knockout mice gait analysis methods were completed. Gait analysis in Prg4-/- mice and knee joint friction studies are ongoing for osteoarthritis investigation. Results: PRG4 suppresses MSU crystal growth (average 8 crystals at 96 hrs, 100 mcg/ml) as compared to BSM (average 231 crystals at 96 hrs, 100 mcg/ml). Also, pre-formed MSU crystals biofouled with rhPRG4 suppress CXCL and IL1β expression. In vivo, Prg4 conditional mice injected with MSU crystals in the presence (swing time, 0.0795 s) and absence (swing time, 0.0468 s) of lubricin production showed detectable differences in gait analysis. Conclusions: These data are important due to the rising cases of gout and the necessity for alternative treatments in both acute and chronic cases. Future work will focus on understanding the mechanism of action between PRG4 and MSU crystal formation.
