It has two main subtypes: Non-Small Cell Lung Cancer (NSCLC), which comprises 85% of cases, and Small Cell Lung Cancer (SCLC). Major issues include delays in diagnosis and treatment. A personalized treatment strategy, particularly through Functional Precision Medicine (FPM), is essential, focusing on heterogeneity and sample size limitations. Strategies like spheroid cultures and Optical Coherence Tomography (OCT) can address these challenges. Methods: Aim 1: focuses on optimizing the spheroid development of A549 by comparing different growth media (DMEM, RPMI, and F12k) and seeding densities (50 x 10³, 100 x 10³, 200 x 10³, and 400 x 10³) to achieve optimal cell growth. For Aim 2: utilizing the best spheroid development from Aim 1, a drug testing experiment was conducted with drugs (Cisplatin, Sotorasib, and Afatinib) across dosage ranges of (0 µM, 0.7 µM, 7 µM, and 35 µM). Results: Aim 1: A549 spheroid development optimization revealed through morphological images, ATP analysis, and Live/Dead analysis that the most optimal growth media and seeding density were RPMI at a seeding density of 50k. Aim 2: The drug testing comparison study presented unexpected results. Afatinib was the only drug to show a significant effect through ATP analysis, while Cisplatin and Sotorasib demonstrated no statistically significant effects. Conclusion: In conclusion, optimal A549 spheroid development using microtissue models was successful. Further testing for drug studies is necessary due to inconclusive ATP data.
