Abstract
The use of RNA for diagnosis and treatment becomes more rampant with each passing day. One of the main techniques used to analyze RNA is Next Generation Sequencing (NGS). NGS RNA-Seq workflows are known to be labor-intensive and at times difficult to manage because of the nature of the enzymes and RNA. This thesis sets out to find comprehensive ways to automate an array of NGS Library Preparation assays with an added focus on RNA-Seq workflows. The final automated workflows generated for each of the assays were able to produce standardized final libraries comparable to manual samples (positive controls). This was determined by doing quality control and analyzing an array of sequencing parameters for each of the final libraries. To achieve these results, several iterations of the process were tested. Parameters such as incubation times, reagent volumes, mixing techniques, and temperature were iterated until we obtained the results closest to those of the already validated manual ones. Lastly, challenges often encountered by smaller research centers with limited resources were taken into account to make these processes accessible to all.
