  <mods:abstract>Gold nanorods (AuNRs) enable remote neuronal activation via photothermal effects, but their systemic safety after ocular delivery is not well defined. We evaluated systemic toxicity in mice for 32 days after intravitreal injection of three AuNR formulations: 25-nm Thy-1–conjugated AuNRs, 10-nm Thy-1–conjugated AuNRs (size effect), and 25-nm bare AuNRs (surface chemistry effect), with phosphate-buffered saline as control. Hematology, serum biochemistry, and body weight were measured at 2, 8, and 32 days post-injection. Thy-1–conjugated AuNRs produced no significant changes in serum indices or blood counts versus controls, indicating no detectable systemic toxicity. In contrast, bare AuNRs increased uric acid by 23% (p = 0.0018), suggesting systemic effects when targeting is absent. Age and sex influenced several baseline hematologic and biochemical measures. Overall, Thy-1–conjugated AuNRs appear systemically safe through 32 days, underscoring the value of targeted nanoparticle design to reduce toxicity risk.</mods:abstract>
