- While 2D in vitro cell culture models have been used over the decades, 3D cell culture technique is becoming increasingly important as it better replicates the in vivo tumor environment. However, methods of assessing chemosensitivity in 3D culture tissues have been relatively less developed. Traditional assays are limited in probing 3D culture tissues deep inside or require cell disruption that limits the premise for a longitudinal drug test. Optical Coherence Tomography (OCT) has been tested for its potential as a means to assess cell viability in 3D culture tissues: it may enable label-free, non-destructive, and high-throughput assessment but has not been fully demonstrated. MCF7 breast cancer cell line has been widely used to form spheroids as a 3D in-vitro platform for drug testing. Here, we created MCF7 spheroids using the Microtissues™ scaffold-free platform and used OCT to acquire their three-dimensional images longitudinally. This study investigates how various OCT metrics capture MCF7 responses to Paclitaxel treatment, in both dose- and time-dependent manners. This label-free assessment was compared to the gold-standard traditional assay, Live/Dead®.
