- Signals from fluorescent widefield calcium imaging of neural activity are susceptible to contamination and artifacts from vasculature in the brain. Hemoglobin is a strong absorber of fluorescent signals, particularly those emitted by GCaMP. As a result of neurovascular coupling, neural activity can cause oxygenated and deoxygenated hemoglobin changes that interfere with the detection of a fluorescent response. This thesis examines a method by which hemodynamic contamination can be corrected for using a combination approach of both widefield fluorescent imaging and intrinsic optical signaling (IOS) imaging, an imaging method that can be used to track hemodynamics associated with neural activity. The first part of this thesis discusses the development of a piezoelectric whisker stimulation system that can stimulate the animal whisker to cause strong neural activity in the brain. Then the instrumentation of an IOS imaging system and analysis methods using two wavelengths of light to calculate hemoglobin concentrations are also examined. Finally, traditional IOS methods are advanced by including a third wavelength of light into the analysis methods. Ultimately, compared to the two wavelength analysis, the three wavelength analysis method was found to provide a more accurate calculation of deoxygenated hemoglobin concentration changes by reducing the dominating effect of oxygenated hemoglobin concentrations in the data. In the future, these calculations can be used to correct fluorescent data, and eventually used to prove the efficacy of a novel retinal prosthetic.
