- Abstract:
- Despite the existence of metabolomics as a field for nearly two decades, challenges of sample preparation limit the large-scale application and throughput of targeted metabolomics for tier-2 diagnostics. In-depth understanding of the sample microenvironment can enable workflow simplification of clinical samples, especially micro-scale samples, or microsamples, and help us develop technologies for immediate clinical translation. This includes addressing the issues of sample volumes, sample transport, and the cost associated with assay development. With our understanding of the current state of clinical metabolomics in the United States, and the challenges associated with microsample analyte recovery for liquid chromatography tandem mass spectrometry quantification, we have performed several analyses leveraging this technology to advance its capabilities for clinical diagnostics. Combining engineering principles with the knowledge of clinical chemistry, we present innovations that can alter the landscape of sample preparation methods and their subsequent automatability for patient sample testing.
