Development of an Analytical Method for Polymeric Nanoparticle Quantification in Serum by Fourier Transform-Infrared Spectroscopy

Abstract:
This work aims to develop a method of using Fourier-Transform Infrared Spectroscopy (FT-IR) for quantification of polymer levels in biological samples, ex vivo. A method of producing a calibration curve for Sprague Dawley rat serum and PLA nanoparticles (NP) was developed, and tested for reproducibility and feasibility. Individual samples were prepared from stock solutions of potassium bromide (KBr), the biological sample (rat serum), and Poly-L-Lactic Acid (8 kDa) nanoparticles. For each interferogram at a given concentration of PLA, ratios between the absorbance peaks near 1650 and 1750 〖cm〗^(-1 )were recorded. Linearity was shown within 2.5-10% of PLA: serum (w/w) and this relationship was used to calculate NP absorption in vivo. The results show promise in the method’s ability to detect and quantify biodegradable polymers in serum.
